Day 2 — Saturday May 30, 2026

Source: synthesized from public coverage and company releases (see Sources). The Plenary headliners (LBA1–5) land Sunday; Saturday's science was carried by the frontMIND lymphoma readout, the ceremonial Opening Session, and a clutch of practice-shaping breast and prostate datasets.

The 2026 ASCO Annual Meeting moved into its first full science day in Chicago. The marquee readout was frontMIND — the first positive frontline trial in a generation to beat R-CHOP in high-risk aggressive B-cell lymphoma — published simultaneously in The Lancet. The ceremonial spine of the day was the Opening Session, where President Eric J. Small, MD, FASCO delivered his address on the meeting's theme, "The Science and Practice of Translation: Improving Cancer Outcomes Worldwide," alongside an address from NCI director W. Anthony Letai, MD, PhD and the David Karnofsky Award Lecture from Nancy E. Davidson, MD, FASCO. Around the ceremony, breast and GU oral sessions delivered new SERD, biparatopic-bispecific, and de-escalation data.

Hematology — frontMIND headlines the day

frontMIND was the day's defining readout: adding tafasitamab + lenalidomide to R-CHOP cut the risk of progression or death by 25% vs R-CHOP alone in previously untreated high-intermediate/high-risk DLBCL or HGBL (HR 0.75, 95% CI 0.59–0.96, P=.019; 24-mo PFS 71.1% vs 62.9%; N=899). Interim OS was not yet significant (HR 0.85, P=.27). Investigators framed the triplet as a potential new standard — only the second phase 3 in ~25 years to improve a primary efficacy endpoint over R-CHOP in newly diagnosed DLBCL. Simultaneous publication in The Lancet.

Breast — adjuvant SERD and a biparatopic bispecific

The oral SERD story continued with lidERA: a Saturday menopausal-status subgroup analysis confirmed that adjuvant giredestrant improves invasive disease-free survival in ER+/HER2− early breast cancer regardless of menopausal status (overall iDFS HR 0.70; premenopausal HR 0.65, postmenopausal HR 0.74; 3-yr iDFS 92.4% vs 89.6%; N=4,170) — building on the 30% recurrence reduction first shared in late 2025.

KN026-004 (Neo-Healer) brought a HER2 biparatopic bispecific to the neoadjuvant setting (LBA660): anbenitamab + nab-docetaxel ± carboplatin beat THP on total pathologic complete response (tpCR 62.4% vs 51.2%; breast pCR 64.6% vs 55.0%; one-sided P=.0099; N~520).

A NATALEE translational analysis showed adjuvant ribociclib iDFS benefit holds across all PAM50 subtypes (HR range 0.42–0.77, no subtype×treatment interaction across 3,022 samples), hinting at gene-expression-guided risk refinement rather than patient selection.

Genitourinary — de-escalating in exceptional responders

A-DREAM (Alliance A032101) tested planned ADT+ARPI interruption in exceptional mHSPC responders: of 78 patients suspending therapy, 57.7% (45) remained treatment-free at 18 months and 66.7% (52) recovered testosterone (median 24.5 months off treatment) — supporting the feasibility of intermittent therapy and treatment holidays in deep responders.

Other oral data reported Saturday

  • Olverembatinib 2L — second-line olverembatinib in CML-CP (N=42) reached 91.3% CCyR and 60.9% MMR by cycle 24.
  • PRIMAVERA — oral PRMT5 inhibitor AZD3470 in R/R classic Hodgkin lymphoma: 44% ORR overall, 63% ORR / 44% CR at 600 mg.
  • Satri-cel (CT041 Cohort 3) — Claudin18.2 CAR-T sequential after 1L gastric/GEJ: ORR 100% in evaluable patients, median PFS-from-1L 20.9 mo at >4.5 yr follow-up.
