Abstract: 6505 · Session: Oral presentation (Hematologic Malignancies—Leukemia), Tuesday, Jun 2, 2026, 9:45 am–12:45 pm CT · Presenter: not yet disclosed (Syndax/investigators) · Embargo: through presentation, Jun 2, 2026

At a glance

  • Sponsor: Syndax Pharmaceuticals
  • PI / presenter: Not yet disclosed in pre-meeting release
  • NCT ID: Not specified in announcement (Syndax revumenib post-HSCT maintenance program; related post-transplant study NCT06575296)
  • Phase: Investigator/early-phase maintenance experience (observational + historical-control comparison)
  • Design: Revumenib as maintenance therapy following allogeneic hematopoietic stem cell transplantation (allo-HSCT), with comparison to a historical cohort of the same genetic subtypes treated before revumenib

  • Line: Post-allo-HSCT maintenance

Headline result

Syndax reported in its pre-meeting announcement that the oral presentation will describe favorable overall survival and relapse outcomes among 21 adults and children with KMT2Ar, NPM1m, or NUP98r acute leukemia who received revumenib as post-transplant maintenance, benchmarked against a historical cohort of the same genetic subtypes treated before revumenib became available. Specific OS, relapse-rate, HR, and p-value figures were not released pre-meeting and are embargoed until the Jun 2, 2026 presentation (per Syndax topline, May 21, 2026).

Mechanism / class

Revumenib (Revuforj) is an oral, first-in-class menin inhibitor that disrupts the menin–KMT2A protein interaction driving leukemic transcriptional programs in KMT2A-rearranged and NPM1-mutant leukemias. FDA-approved for relapsed/refractory acute leukemia with a KMT2A translocation (adult and pediatric ≥1 year) and for relapsed/refractory AML with a susceptible NPM1 mutation (adult and pediatric ≥1 year with no satisfactory alternative).

Discussant takeaway

Not reported pre-meeting; omit pending presentation.

Practice-changing?

Pending. Post-transplant maintenance is a genuine unmet need in menin-dependent acute leukemias, but this is a small (N=21), historically-controlled experience rather than a randomized trial. Hypothesis-generating; the randomized phase 3 newly-diagnosed programs (EVOLVE-2, REVEAL-ND) will drive any practice change.
